Theoretically speaking, I would imagine that about 10 tons of weight would squish the tire down until it touched the rim. Either the tire would explode or the pressure would go up a bit.

Under reasonable loads, I would expect the volume to remain near constant. Therfore, the pressure would remain unchanged.

The biggest cause of pressure change is likely to be increased temperature as you drive down the road.
